On the front of the VLOM there are two plugins, one for oil pressure and the other must be for activating the 4 cyl mode. If that plug was disconnected would that disable the 4 cyl mode? Maybe it wouldn't need to be tuned out or use the OBD plugin disabler.

It would disable the AFM, But at a big cost without a tune. Your vehicle would immediately enter limp home mode and disable you from operating the throttle. It would also throw you a check engine light among others.
Since you say it is located at the front of the engine you must have one of the newer LT based engines. With that in mind you will definitely want to have it flashed. You will also definitely want to add the VLOM plug. The newer LT direct injection engines are known for having a leaky VLOM. What they end up doing is leaking into the oil tower where it adds pressure to the AFM portion of the lifter when it's not supposed to. This of course causes the locks to bind and fail leading to a collapsed/stuck AFM lifter. The newer LT-based stuff is also known for bending pushrods because of factory interference that is caused from the angle of the valvetrain versus the diameter of the pushrod. I sell a smaller diameter pushrod to solve for this problem.
